ﻫﺪف از اﻧﺠﺎم ﭘﮋوﻫﺶ ﺣﺎﺿﺮ ، ﺗﻌﯿﯿﻦ ﺗﺄﺛﯿﺮ ﺗﻤﺮﯾﻦ ﻫﻮازي ﺑﺎ ﺷﺪت ﻣﺘﻮﺳﻂ و زﯾﺎد ﺑﺮ ﺑﯿﺎن ژنﻫﺎيBAX و BCL2 ﻣﯿﻮﮐﺎرد ﻣﻮشﻫﺎي ﺻﺤﺮاﯾﯽ ﻧﺮ ﭘﺲ از ﮐﻢﺧﻮﻧﯽ و ﺧﻮنرﺳﺎﻧﯽ ﻣﺠﺪد ﺑﻮد. در اﯾﻦ ﭘﮋوﻫﺶ از26 ﺳﺮ ﻣﻮش ﺻﺤﺮاﯾﯽ وﯾﺴﺘﺎر دوﻣﺎﻫﻪ ﺑﺎ وزن ﺗﻘﺮﯾﺒﯽ25/25 ± 225/25 ﮔﺮم اﺳﺘﻔﺎده ﺷﺪ. ﺑﺮاي ﺑﺮرﺳﯽ اﯾﺠﺎد ﺳﮑﺘﮥ ﺣﺎﺻﻞ از ﺗﺰرﯾﻖ اﯾﺰوﭘﺮﻧﺎﻟﯿﻦ ، ﻣﻄﺎﻟﻌﻪ اي راﻫﻨﻤﺎ در ﮔﺮوه ﺗﺠﺮﺑﯽ ﺑﺮاي ﮔﺮوه ﺳﮑﺘﻪ )ﺗﻌﺪاد = ﻫﻔﺖ( و ﮔﺮوه ﮐﻨﺘﺮل ﺗﻌﺪاد = ﻫﻔﺖ( اﻧﺠﺎم ﺷﺪ و ﺳﮑﺘﮥ ﺑﻄﻦ ﭼﭗ ﭘﺲ از ﺗﺰرﯾﻖ اﯾﺰوﭘﺮﻧﺎﻟﯿﻦ ﺑﺎ ﺗﮑﻨﯿﮏ ﺗﺮيﮐﺮوﻣﺎﺳﻮن ﺗﺄﯾﯿﺪ ﺷﺪ ﺳﭙﺲ ، ﻣﻮشﻫﺎي ﮔﺮوه ﭘﮋوﻫﺶ ﺑﺮاي اﻧﺪازه ﮔﯿﺮي ﺑﯿﺎن ژن ﻫﺎ ﺑﻪ ﺳﻪ ﮔﺮوه ﺗﻤﺮﯾﻦ ﻫﻮازي ﺑﺎ ﺷﺪت ﻣﺘﻮﺳﻂ )ﺗﻌﺪاد ﭼﻬﺎر( ، ﺷﺪت زﯾﺎد )ﺗﻌﺪاد = ﭼﻬﺎر( و ﮐﻨﺘﺮل )ﺗﻌﺪاد = ﭼﻬﺎر( ﺗﻘﺴﯿﻢ ﺷﺪﻧﺪ. ﻣﻮشﻫﺎي ﮔﺮوه ﭘﮋوﻫﺶ ﺑﻪﻣﺪت ﯾﮏ ﻫﻔﺘﻪ ﺑﺎ دوﯾﺪن روي ﻧﻮار ﮔﺮدان آﺷﻨﺎ ﺷﺪﻧﺪ و ﺗﻤﺮﯾﻦ ﻫﻮازي ﯾﮏﻣﺎﻫﻪ را ﺑﺎ ﺷﺪت ﻣﺘﻮﺳﻂ و زﯾﺎد ﺑﺎ ﺗﻮاﺗﺮ ﺳﻪ ﺟﻠﺴﻪ در ﻫﻔﺘﻪ اﺟﺮا ﮐﺮدﻧﺪ. ﭘﺲ از دو روز اﺳﺘﺮاﺣﺖ، ﺑﺎ ﻓﺎﺻﻠﮥ 24 ﺳﺎﻋﺖ ﺑﻪ ﻣﻮشﻫﺎ اﯾﺰوﭘﺮﻧﺎﻟﯿﻦ ﺗﺰرﯾﻖ ﺷﺪ ، اﻣﺎ ﻪ ﻣﻮشﻫﺎي ﮔﺮوه ﮐﻨﺘﺮل ﻓﻘﻂ اﯾﺰوﭘﺮﻧﺎﻟﯿﻦ ﺗﺰرﯾﻖ ﺷﺪ. از ﻓﺮﻣﻮلct ∆∆-2 ﺑﺮاي اﻧﺪازه ﮔﯿﺮي ﺑﯿﺎن ژن BAX و BCL2 و از آزﻣﻮن آﻣﺎري ﺗﯽ ﻣﺴﺘﻘﻞ ﺑﺮاي ﻣﻘﺎﯾﺴﮥ دو ﮔﺮوه ﺳﮑﺘﻪ و ﮐﻨﺘﺮل در ﻣﻄﺎﻟﻌﮥ راﻫﻨﻤﺎ و از آزﻣﻮن آﻧﻮا ﺑﺮاي ﻣﻘﺎﯾﺴﮥ ﮔﺮوهﻫﺎ اﺳﺘﻔﺎده ﺷﺪ. ﻧﺘﺎﯾﺞ ﭘﮋوﻫﺶ ﻧﺸﺎن داد ﮐﻪ ﺗﻤﺮﯾﻦ ﻫﻮازي ﻣﺘﻮﺳﻂ و زﯾﺎد ﻣﯽﺗﻮا ﻧﺪ ﺑﯿﺎن ژن BAX و BCL2 را ﺑﻪﺗﺮﺗﯿﺐ ﮐﺎﻫﺶ و اﻓﺰاﯾﺶ دﻫﺪ ، اﻣﺎ اﯾﻦ ﺗﻐﯿﯿﺮات در ﺗﻤﺮﯾﻦ ﻫﻮازي ﺑﺎ ﺷﺪت ﻣﺘﻮﺳﻂ و زﯾﺎد ﻣﻌﻨﺎدار ﻧﺒﻮد؛ ﺑﻨﺎﺑﺮاﯾﻦ ، ﺑﻪﻧﻈﺮ ﻣﯽرﺳﺪ اﯾﻦ ﺷﺪت و ﻣﺪت زﻣﺎن ﺗﻤﺮﯾﻦ ﻧﻤﯽﺗﻮاﻧﺪ ﻣﺴﯿﺮﻫﺎي آﻧﺘﯽآﭘﻮﭘﺘﻮزي ﺣﺎﺻﻞ از ورزش را ﺑﺮاي ﺑﻪﺣﺪاﻗﻞرﺳﺎﻧﺪن ﻋﻮارض ﺳﮑﺘﻪ ﻓﻌﺎل ﮐﻨﺪ.
چكيده لاتين :
The present research aims to examine the effect of aerobic exercise with medium and high intensity on the gene expression of Bax and Bcl-2 markers in rat myocard after ischemic-reperfusion by isoprenaline. 26 male Wistar rats were used weighing approximately 225.25±25.25 g with two to three months old; Then pilot study with stroke group (n = 7) and control Group (n = 7) was knock to examine of isopernalin induced-infarct and the infarct of left ventricle were confirmed in isopernalin into normal groups used Masson-trichrom teqnique. Then rats were divided into three groups to Measure gene expression: control (n=4), aerobic exercise with middle (n=4) and many intensity (n=4). The rats in research group were introduced to treadmill for a week, then the rats were exposed medium and high intensity aerobic training with frequency of three times weekly for one month. After two days of rest, rats were injected with isoprenaline within 24 hours; but control group were injected only with isopernaline in the end of one month. The formula 2-ΔΔct were used to measure the gene expression of BAX and BCL2, and independent t-test was used to compare the two groups of stroke and control in the pilot study and ANOVA were used to compare the groups.The results of study showed that aerobic exercise with medium and high intensity could reduce and increase the gene expression of BAX and BCL2, but these changes were not significant. Therefore, it seems that this intensity and period of training can not active anti-apoptotic routes of exercise to minimize the effects of stroke.
