ﺳﺎﺑﻘﻪ و ﻫﺪف: ﻫﯿ ﺴ ﺘﻮن دا ﺳ ﺘﯿﻼزﻫﺎ-4 )HDAC4( ﮐﻪ ﻓ ﺴﻔﻮرﯾﻠﻪ ﺷﺪن آنﻫﺎ ﺑﻪو ﺳﯿﻠﻪ ﮐﯿﻨﺎز واﺑ ﺴﺘﻪ ﺑﻪ ﮐﻠ ﺴﯿﻢ ﮐﺎﻟﻤﻮدوﻟﯿﻦ )CaMKII( ﺻﻮرت ﻣﯽﮔﯿﺮد، ﻧﻘﺶ ﻣﻬﻤﯽ در رﺷﺪ و ﻧﻤﻮ ﺑﺎﻓﺖ ﻗﻠﺐ دارد. از اﯾﻦرو ﻫﺪف از ﻣﻄﺎﻟﻌﻪ ﺣﺎﺿﺮ، ﺑﺮرﺳﯽ اﺛﺮ ﻓﻌﺎﻟﯿﺖ ورزﺷﯽ ﺑﺎ ﺷﺪت ﻣﺘﻮﺳﻂ ﺑﺮ ﺑﯿﺎن ژنﻫﺎي HDAC4 و CaMKII در ﻣﯿﻮﮐﺎرد رتﻫﺎي ﻧﺮ ﻧﮋاد وﯾﺴﺘﺎر ﺑﻮد. ﻣﻮاد و روشﻫﺎ: در اﯾﻦ ﻣﻄﺎﻟﻌﻪ ﺗﺠﺮﺑﯽ، 19 ﺳﺮ ﻣﻮش ﺻﺤﺮاﯾﯽ ﻧﺮ ﺑﺎﻟﻎ ﺑﺎ 10 ﻫﻔﺘﻪ ﺳﻦ و وزن 243±8/2 ﮔﺮم ﺑﺮاﺳﺎس ﻫﻤﺴﺎن ﺳﺎزي وزن ﺑﻪ 2 ﮔﺮوه )ﺗﻤﺮﯾﻦ اﺳﺘﻘﺎﻣﺘﯽ =9( و )ﮐﻨﺘﺮل=10( ﺗﻘﺴﯿﻢ ﺷﺪﻧﺪ. ﺑﺮﻧﺎﻣﻪ ﺗﻤﺮﯾﻨﯽ ﺷﺎﻣﻞ 6 ﻫﻔﺘﻪ ﺗﻤﺮﯾﻦ اﺳﺘﻘﺎﻣﺘﯽ، 5 روز در ﻫﻔﺘﻪ ﺑﺎ ﺷﺪت ﻣﺘﻮﺳﻂ ﺑﻮد. 24 ﺳﺎﻋﺖ ﭘﺲ از آﺧﺮﯾﻦ ﺟﻠﺴﻪ ﺗﻤﺮﯾﻦ، ﻧﻤﻮﻧﻪﻫﺎي ﺑﺎﻓﺖ ﻗﻠﺐ ﺑﺮاي اﻧﺪازهﮔﯿﺮي ﺳﻄﻮح ﺑﯿﺎن ژنﻫﺎي HDAC4 و CaMKII ﺑﺎ اﺳﺘﻔﺎده از روش آزﻣﺎﯾﺸــﮕﺎﻫﯽ Real Time PCR ﻣﻮرد ﺑﺮرﺳــﯽ ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﺗﺠﺰﯾﻪ و ﺗﺤﻠﯿﻞ دادهﻫﺎ از ﻃﺮﯾﻖ ﻧﺮماﻓﺰار SPSS وﯾﺮاﯾﺶ 23 و ﺑﺎ اﺳﺘﻔﺎده از آزﻣﻮن ﺗﯽ ﻣﺴﺘﻘﻞ اﻧﺠﺎم ﺷﺪ.
ﻧﺘﺎﯾﺞ: ﯾﺎﻓﺘﻪﻫﺎ ﻧﺸــﺎن داد ﻣﯿﺰان ﺑﯿﺎن ﻫﺮ دو ژن HDAC4 و CaMKII در ﮔﺮوه ﺗﻤﺮﯾﻦ ﺑﻪﻃﻮر ﻣﻌﻨﯽداري ﮐﻤﺘﺮ از ﮔﺮوه ﮐﻨﺘﺮل ﺑﻮده اﺳــﺖ )0/001
چكيده لاتين :
Histone Dacetylases-4 (HDAC4) is phosphorylated by calcium-calmodulindependent
kinase (CaMKII) that plays an important role in cardiac tissue hypertrophy.
Therefore, the present study aimed to investigate the effect of moderate intensity exercise on
HDAC4 and CaMKII genes expression in myocardial Wistar male rats.
Materials and Methods: In this experimental study, 19 adult male rats with 10 weeks of age
and weighing of (243±8.2 g) were divided into 2 groups (endurance training=9) and
(control=10) based on weight matching. Training program consisted of 6 weeks of endurance
training, 5 days per week and moderate intensity. 24 hours after the last training session,
cardiac tissue samples were extracted to measure the expression levels of HDAC4 and
CaMKII by in vitro Real Time PCR. Data were analyzed using SPSS software version 23 and
independent t-test was used.
Results: The results showed that the expression of both HDAC4 and CaMKII genes in the
exercise group was lower than the control group, significantly (P<0.001).
Conclusion: According to the results of the present study, it seems that moderate intensity
endurance training can change some factors related to cardiac hypertrophy tissue of rats and
can be effective in preventing cardiovascular disease.
