چكيده فارسي :
زﻣﯿﻨﻪ و ﻫﺪف ﮐﺎﻧﺎﺑﯿﺲ، ﻣﺎده روان ﮔﺮداﻧﯽ اﺳﺖ ﮐﻪ ﻣﻮرد ﺳﻮءﻣﺼﺮف ﻣﯿﻠﯿﻮن ﻫﺎ ﻧﻔﺮ در دﻧﯿﺎ ﻗﺮار ﻣﯽ ﮔﯿﺮد. ﺑﺎ ﺗﻮﺟﻪ ﺑﻪ ﺷﯿﻮع ﻧﺎﺑﺎروي و ﻣﺼﺮف زﯾﺎد اﯾﻦ ﻣﺎده در ﺑﯿﻦ ﺟﻮاﻧﺎن، ﻣﻄﺎﻟﻌﻪ ﺣﺎﺿﺮ ﺑﺎ ﻫﺪف ﺑﺮرﺳﯽ ﺗﺄﺛﯿﺮ ﻋﺼﺎره ﮐﺎﻧﺎﺑﯿﺲ ﺑﺮ رﺷﺪ ﺳﻠﻮل ﻫﺎي ﺑﻨﯿﺎدي ﻣﺰاﻧﺸﯿﻤﯽ ﻣﺸﺘﻖ از آﻧﺪوﻣﺘﺮ ﻣﻮش ﻫﺎي ﺻﺤﺮاﯾﯽ ﺑﺎﻟﻎ اﻧﺠﺎم ﮔﺮدﯾﺪ. ﻣﻮاد و روشﻫﺎ: در اﯾﻦ ﻣﻄﺎﻟﻌﻪ ﺗﺠﺮﺑﯽ، ﺳﻠﻮﻟﻬﺎي ﺑﻨﯿﺎدي ﻣﺰاﻧﺸﯿﻤﯽ از آﻧﺪوﻣﺘﺮ رﺣﻢ ﻣﻮش ﻫﺎي ﺻﺤﺮاﯾﯽ اﺳﺘﺨﺮاج ﮔﺮدﯾﺪ و ﭘﺲ از ﮐﺸﺖ و ﺗﺄﯾﯿﺪ ﻣﺰاﻧﺸﯿﻤﯽ ﺑﻮدن ﺳﻠﻮل ﻫﺎ ﺑﺎ روش ﻓﻠﻮﺳﺎﯾﺘﻮﻣﺘﺮي و ﺑﺮاﺳﺎس ﺑﯿﺎن ﻣﺎرﮐﺮﻫﺎي CD34و CD90و ﺑﯿﺎن ﻧﺸﺪن ﻣﺎرﮐﺮ CD105، در ﭘﺎﺳﺎژ ﺳﻮم ﮐﺸﺖ ﺳ ﻠﻮﻟﯽ، ﺗﺄﺛﯿﺮ ﮐﺎﻧﺎﺑﯿﺲ در ﻏﻠﻈﺖﻫﺎي 1000 ng/mlو 100 در ﻃﻮل ﻣﺪت 1 اﻟﯽ8 روز ﺑﺮ ﻣﯿﺰان رﺷﺪ اﯾﻦ ﺳﻠﻮل ﻫﺎ ﺗﻮﺳﻂ ﻓﺮﻣﻮل PDT= T×ln𝑙𝑙𝑙𝑙Xe ﻣﺤﺎﺳﺒﻪ ﮔﺮدﯾﺪ و ﻧﺘﺎﯾﺞ ﺑﺎ اﺳﺘﻔﺎده از آزﻣﻮن ﻫﺎي ANOVA و ﺗﻮﮐﯽ آﻧﺎﻟﯿﺰ ﺷﺪﻧﺪ. ﯾﺎﻓﺘﻪﻫﺎ: ﺳﻠﻮل ﻫﺎي ﺟﺪا ﺷﺪه از آﻧﺪوﻣﺘﺮ، 24 ﺳﺎﻋﺖ ﭘﺲ از اﻧﺘﻘﺎل، ﮐﺎﻣﻼً ﺑﻪ ﮐﻒ ﻓﻼﺳﮏ ﮐﺸﺖ ﺳﻠﻮﻟﯽ ﭼﺴﺒﯿﺪﻧﺪ. ﻣﺰاﻧﺸﯿﻤﯽ ﺑﻮدن اﯾﻦ ﺳﻠﻮل ﻫﺎ ﺑﺎ ﺑﯿﺎن ﻣﺎرﮐﺮﻫﺎي ﺳﻄﺤﯽ CD105 ،CD90 و ﺑﯿﺎن ﻧﺸﺪن ﻣﺎرﮐﺮ CD34 ﺗﺄﯾﯿﺪ ﮔﺮدﯾﺪ. ﻧﺘﺎﯾﺞ ﺷﻤﺎرش ﺳﻠﻮﻟﯽ ﻧﯿﺰ ﺣﺎﮐﯽ از رﺷﺪ ﺳﻠﻮل ﻫﺎي ﺗﯿﻤﺎر ﺷﺪه ﺑﺎ ﮐﺎﻧﺎﺑﯿﺲ ﺗﺎ روز ﺳﻮم ﺗﯿﻤﺎر ﻣﺸﺎﺑﻪ ﺑﺎ ﮔﺮوه ﮐﻨﺘﺮل ﺑﻮد و از روز ﭼﻬﺎرم درﮔﺮوه ﺗﯿﻤﺎر ﺑﺎ دوز 100ng/mlﮐﺎﻧﺎﺑﯿﺲ اﻓﺰاﯾﺶ ﻣﻌﻨﺎداري در ﺳﻄﺢ p<0 /05 و در ﮔﺮوه ﺗﯿﻤﺎر ﺑﺎ دوز 1000ng/ml در روز ﺳﻮم ﻧﺴﺒﺖ ﺑﻪ ﮔﺮوه ﮐﻨﺘﺮل ﮐﺎﻫﺶ ﻣﻌﻨﺎدار و از روز ﭼﻬﺎرم اﻓﺰاﯾﺶ ﻣﻌﻨﺎداري در ﺳﻄﺢ p<0 /01 ﻣﺸﺎﻫﺪه ﮔﺮدﯾﺪ. ﻧﺘﯿﺠﻪ ﮔﯿﺮي: ﻧﺘﺎﯾﺞ ﻧﺸﺎن داد ﺳﻠﻮل ﻫﺎي ﺟﺪا ﺷﺪه از آﻧﺪوﻣﺘﺮ، از ﻧﻮع ﺑﻨﯿﺎدي ﻣﺰاﻧﺸﯿﻤﯽ ﺑﻮدﻧﺪ و ﻋﺼﺎره ﮐﺎﻧﺎﺑﯿﺲ اﺣﺘﻤﺎﻻ ًاز ﻃﺮﯾﻖ ﮔﯿﺮﻧﺪهﻫﺎي ﮐﺎﻧﺎﺑﯿﻨﻮﺋﯿﺪي ﺑﺎﻋﺚ ﺗﺤﺮﯾﮏ رﺷﺪ اﯾﻦ ﺳﻠﻮل ﻫﺎ ﻣﯽﮔﺮدد .
چكيده لاتين :
Cannabis is psychoactive substance that is abused by millions of people the world. Due to the high consumption of this substance among young people of reproductive age, the present study was performed to investigate the effect of cannabis on the growth of endometrial mesenchymal stem cells(msc) derived from rats.
Materials and Methods: In this experimental study, msc were extracted from the endometrium of rats and after culturing and confirming the mesenchymal nature of the cells by flowcytometry and by expressingCD34 and CD90 and not expressingCD105 markers,in the third passage of cell culture, the effects of cannabis in concentration of100and1000ng/ml were calculated on the growth of these cells within1to8days by the
PDT=T×𝑙𝑙𝑙𝑙2lnXeXb formula and the results were analyzed using ANOVA and Tuki tests.
Results: Cells isolated from the endometrium adhered to the floor of the cell culture flask24hours after transfer. The mesenchymal nature of these cells was confirmed by the expression of CD90, CD105and non-expression of CD34markers.The results ofcell counts also showed the growth of cells treated with cannabis until the third day of treatment similar to the control group. From the fourth day in the treatment group with a dose of100ng/ml cannabis increased significantly top<0.05and in the treatment group with a dose of1000ng/ml. Third, there was significant decrease compared to the control group and from the fourth day, significant increase was observed atp<0.05.
Conclusion: The results showed that the cells isolated from the endometrium were of the msc, and that cannabis probably stimulated the growth of these cells through cannabinoid receptors.
