چكيده فارسي :
ﻧﻘش ﮔونه ھﺎي ﻓﻌﺎل اﮐﺳﯾژن و آﺳﯾب اﮐﺳﯾداﺗﯾو ﻧﺎﺷﯽ از آنھﺎ در ﺑﺳﯾﺎري از ﭘروسه ھﺎي ﭘﺎﺗوﻟوژﯾﮏ ﻣﺷﺧص ﮔردﯾده اﺳت . ﺑﻧزوآﻟﻔﺎﭘﺎﯾرن ﯾﮏ ﺗرﮐﯾب ھﯾدروﮐرﺑﻧﮫ ﺣﻠﻘوي ﭼﻧد زﻧﺟﯾره ﻣﻌطر ﺑﺎ وزن ﻣوﻟﻛوﻟﻲ ﺑﺎﻻ ﺑوده ﮐﮫ اﺛرات ﺳرطﺎنزاﯾﯽ ﺑﺎﻻﯾﻲ دارد. ﻟذا ھدف اﯾن ﻣطﺎﻟﻌﮫ در ﺳﺎل 1397ﺑررﺳﻲ اﺛر ﺑﻧزوآﻟﻔﺎﭘﺎ ﯾرن ﺑر ﺷﺎﺧصھﺎي اﺳﺗرس اﮐﺳ ﯾداﺗﯾو در ﺑﺎﻓت كليه در ﻣﺎھﯽ ھﺎﻣور ﻣﻌﻣوﻟﯽ اﺳت. 25ﻋدد ﻣﺎھﻲ ﺑﺎ ﻣﯾﺎﻧﮕﯾن وزﻧﯽ 400±50ﮔرم ﭘس از اﻧﺗﻘﺎل ﺑﮫ ﺗﺎﻧﮏھﺎي آب 300ﻟﯾﺗري ﺑﮫ ﻣدت 7 روز در ﭘﻧﺞ ﮔروه آزﻣﺎﯾﺷﯽ ﺷﺎﻣل ﮔروه ﮐﻧﺗرل، ﮔروه ﺷﺎھد )12 ﻣﯾﮑرو ﻟﯾﺗر ﺣﻼل دي ﻣﺗﯾل ﺳوﻟﻔواﮐﺳﯾد( و ﺳﮫ ﮔروه ﺗﯾﻣﺎر ﺑﻧزوآﻟﻔﺎﭘﺎ ﯾرن ﺑﺎ ﻏﻠظت ﮐم )0/5 ﻣﯾﻛروﮔرم در ﻟﯾﺗر(، ﻏﻠظت ﻣﺗوﺳط ) 1/5ﻣﯾﻛروﮔرم در ﻟﯾﺗر( و ﻏﻠظت ﺑﺎﻻ ) 5ﻣﯾﻛروﮔرم در ﻟﯾﺗر( ﺑﮫ ﻣﺎھﯾﺎن ﺗزرﯾﻖ ﺷد و ﭘس از 7 روز ﺑﺎﻓت ﮐﻠ ﯾﮫ ﺟﮭت ﺑررﺳﯽ ﺗﺳت ھﺎي آﻧﺗﯽ اﮐﺳﯾداﻧﯽ اﺳﺗﺧراج ﮔردﯾد. ﺑراي ﺳﻧﺟش اﺳﺗرس اﮐﺳﯾداﺗﯾو، از ﺳﻧﺟش ﻏﻠظت ﻣﺎﻟون دي آﻟد ﺋﯾد )MDA( و ﻣﯾزان ﺗﯾول اﺳﺗﻔﺎده ﺷد . ﻧﺗﺎﯾﺞ ﺑﺎ اﺳﺗﻔﺎده از آزﻣونھﺎي آﻣﺎري آﻧﺎﻟﯾز وارﯾﺎﻧس ﯾ ﮏطرﻓﮫ و آزﻣون ﺗوﮐﯽ ﺗﺟزﯾﮫوﺗﺣﻠﯾل ﺷدﻧد. ﺳطﺢ ﻣﻌﻧﯽداري، P<0/05در ﻧظر ﮔرﻓﺗﮫ ﺷد. ﺗﻔﺎوت ﻣﻌﻧﯽداري ﻣﯾﺎن ﻣﯾزان ﻣﺎﻟون دي آﻟدﺋﯾد و ﻧﯾز ﻏﻠظت ﺗﯾول ﺑﯾن ﮔروه ھﺎي ﮐﻧﺗرل و ﺷﺎھد در ﺑﺎﻓت ﮐﻠﯾﮫ ﻣﺷﺎھده ﻧﺷد. اﻓزا ﯾش ﻣﻌﻧﯽداري ﺑﯾن ﻣﯾزان ﻓﻌﺎﻟﯾت MDAﮐﻠﯾوي در ﻣﺎھﯾﺎﻧﻲ ﮐﮫ در ﻣﻌرض 0/5، 1/5، 5ﻧﺎﻧوﮔرم در ﻟﯾﺗر ﺑﻧزوآﻟﻔﺎﭘﺎﯾرن ﻗرارﮔرﻓﺗﮫ ﺑودﻧد ﻧﺳﺑت ﺑﮫ ﮔروه ﮐﻧﺗرل ﻣﺷﺎھده ﺷد )P<0/001(. از طرﻓﯽ ﻣﯾزان ﺗﯾول در اﯾن ﻏﻠظت ھﺎ ﻧﺳﺑت ﺑﮫ ﮔروه ﮐﻧﺗرل ﺑﮫ طور ﻣﻌﻧﯽدار ﮐﺎھش ﯾ ﺎﻓت )P<0/001(. ﻣﻘﺎدﯾر ﺑﮫدﺳتآﻣده در اﯾن ﻣطﺎﻟﻌﮫ ﻣﯽﺗواﻧد به ﻋﻧوان ﻣﻌﯾﺎري ﺟﮭت ارزﯾﺎﺑﯽ اﺳﺗرس اﮐﺳﯾداﺗﯾو در ﺑﺎﻓت ﮐﻠﯾﮫ ﻣﺎھﯽ ھﺎﻣور در ﻣﻌرض ﺑﻧزوآﻟﻔﺎﭘﺎﯾرن و ھﻣﭼﻧﯾن ﻣﺎﻧﯾﺗورﯾﻧﮓ آﻟودﮔﯽ ھﺎي ﻣﺣﯾطﯽ اﮐوﺳﯾﺳﺗم ھﺎي آﺑﯽ ﻣورداﺳﺗﻔﺎده ﻗرار ﮔﯾرد.
چكيده لاتين :
The role of reactive oxygen species and their oxidative damage in many pathological processes have been identified. Benzo-α-Pyren is a high molecular weight multi-chain aromatic cyclic hydrocarbon compound with high carcinogenic effects. Therefore, the aim of this study was to investigate the effect of Benzo-α-Pyren on oxidative stress indices in renal tissue in common grouper. 25 Epinephelus coioides were transferred to 300liter water tanks for 7 days in five experimental groups including control group, sham group (12 μl of dimethyl sulfoxide solution) and Three treatment groups Benzo-α-Pyren with low concentration (0.5 μg / l), medium concentration (5). (1 μg / L) and high concentrations (5 μg / L) were injected into fish. After 7 days, kidney tissue was extracted for antioxidant tests. Malondialdehyde (MDA) and thiol levels were used to measure oxidative stress. The results were analyzed using one-way ANOVA and Tukey tests. Significance level was considered P < 0.05. No significant difference was observed between malondialdehyde content and thiol concentration between control and Sham group in kidney tissue. There was a significant increase in the activity of renal MDA in fish exposed to 0.5, 1.5, 5 ng / l Benzo-α-Pyren compared to the control group (P< 0.001). On the other hand, the amount of thiol in these concentrations decreased significantly compared to the control group (P < 0.001). No significant difference was observed in the tissue of all fish of control and control groups of solvent malondialdehyde (MDA) and thiol levels. The values obtained in this study can be used as a criterion for assessing oxidative stress in the tissues of kidneyfish exposed to benzoalphapyrene as well as monitoring the environmental pollution of aquatic ecosystems.
