معرفي گونه‌ي جديدي از جنس Sphaerulina در ايران

New species of Sphaerulina on Populus from Iran

در راستاي مطالعات تاكسونوميك قارچ­هاي سپتوريا و شبه سپتوريا در ايران، گونه­ي جديدي از جنس Sphaerulina روي ميزبان Populus sp. بر اساس توالي يابي ناحيه ITS از rDNA و بررسي­ هاي مرفولوژيك تحت نام Sphaerulina osipyaniae sp. nov. Azimi توصيف شد. مقايسه توالي نمونه مورد مطالعه با توالي Mycosphaerella populi (=Sphaerulina frondicola) از بانك ژن نشان داد كه گونه Sphaerulina روي Populus در ايران گونه­ ي جديدي براي اين جنس است. لكه ­ها در هر دو سطح رويي و زيري برگ، گرد تا كشيده به رنگ خاكستري روشن با حاشيه مشخص به رنگ قهوه­اي تيره، با قطر 5-2 ميلي­متر بودند. لكه ­ها گاهي به­ هم پيوسته و لكه ­هايي نامنظم با اندازه بزرگ‌تر را تشكيل مي­دادند. كنيديوما از نوع پيكنيديوم، نسبتاً گرد، به‌رنگ قهوه­اي تيره تا سياه، در لكه­هاي هر دو سطح رويي و زيري برگ بصورت جدا از ­هم و پراكنده تشكيل شده بودند. قطر پيكنيديوم­ها (200-150) 160 ميكرومتر و داراي دهانه منفرد انتهايي بودند. كنيديوم ­ها داسي شكل، برخي در انتها داراي خميدگي، نيمه شفاف، داراي يك بند، با قاعده گرد و در راس گرد تا كمي باريك شده بودند. طول كنيديوم ­ها 46-35 ميكرومتر و عرض آن 4-3 ميكرومتر اندازه ­گيري شد.

In the framework of study on Septoria and Septoria-like species in Iran, a new species Sphaerulina osypianiae sp. nov. on Populus sp. is described using morphological and rDNA ITS sequencing data. DNA extraction, amplification and sequencing were performed using methods and protocols mentioned by Azimi (2011). The resulting 511-bp sequence obtained from isolate 52935 was deposited in GenBank. The sequence shared 99% identity with Mycosphaerella populi (=Sphaerulina frondicola) sequences available in GenBank (see table 1 for identities). NJ tree generated using TreeView Display Option in NCBI BLAST, supported Sphaerulina species on Populus from Iran as a distinct species from all other Sphaerulina species sequences on Populus.
