Introduction: Caffeine is a nature alkaloid which is found in coffee, tea and cola containing beverages. It is metabolized by cytochrome P-450 enzyme complex in the liver. Since highʹ consumption of caffeine in pregnant women may be induced side effects on the fetal, inthis study we compared the effects of caffeine administration in pregnant C57BL/6J mice with and without induction of maternal metabolism by phenobarbital.
Materials and Methods: In this study 60 pregnant mice C57BL/6J were used. Caffeine was administrated orally or intraperitonaly. In both parts of study three separate groups of animals were used as follows: contol (group 1), caffeine alone (group 2) and caffeine plus phenobarbital (group 3). On day 9 of gestation, groups 1, 2 and 3 received distilled water, caffeine (80 mg/kg) and caffeine (80 mg/kg) + phenobarbital (80 mg/kg), respectively, by oral or IP administrations. On days 11 and 12 of gestation, caffeine (200 mg/kg, IP) was administered to groups 2 and 3. and contorl group received distilled water. On day 17 of gestation, all dams were sacrificed and fetuses were removed, weighed and examined for external hematoma.
Results: Results (weight of fetuses) showed significant differences between caffeine and phenobarbital plus caffein groups in both oral or IP administration conditions (P<0.05). No other findings were statistically significant. Results of external hematoma showed sigificnat difference between control and phenobarbital plus caffein in both conditions. Also differeces between caffein groups ( weight and external hematoma) in both administration conditions were sinificant (P<0.05).
Conclusion: Data indicated that phenobarbital at least dosage of 80 mg/kg abolished the toxic effects of caffeine on fetuses weight.
