بررسي ارتباط بين بروز پروتيين هاي حاصل از ژن هاي P61 ، P35 ، يي كادهرين و بتاكاتينن با عوامل پيش آگهي

DETERMINATION OF CORRELATION OF THE EXPRESSION OF P53,P16, E-CADHERIN an‎d BETA-CATENIN PROTEINS WITH PROGNOSTIC FACTORS IN 50 CASES OF GASTRIC ADENOCARCINOMA BY IMMUNOHISTOCHEMISTRY an‎d TISSUE ARRAY

Background & Aims: Grastric adenocarcinoma is one of the most frequent malignancies. Regrettably, early diagnosis and prediction of response to treatment (chemotherapy or surgery), survival and prognosis of diseas are chaotic problems in regard to this malignancy. - in recent years, many genes were introduced which correlation of their expression with survival and prognosis have suggested, which the most important of them are genes that producing proteins such as P53, Pic,, P21,E-cadherin and Beta Catenin. -The main objection of this study is evaluating the expression of E-cadherin, genes in the cells of adenocarcinoma of stomach by IHC & Tissue array techniques. Materials & Methods: Fifty paraffin blocks of gastric adenocarcinoma from the file of pathology department of Shariati hospital were collected and in selected slides, the appropriate regions for IHC staining were marked with punch instrument (diameter of 4mm), selected regions of each block have removed and then these specimens with their own number, were transferred to tissue array block.Each tissue array block has 10 specimens of gastric adenocarcinoma and 2 built-in control specimens (including one specimen of normal gastric epithelium and another specime of invasive ductal carcinoma of breast). After sectioning, H&E staining and IHC study for aforementioned markers was performed. Results: There is positive statistical correlation between expression of BetaCatenin & P16 with presence of metastasis to regional lymph nodes (p<0.05). Tere is also statistical correlation between P53 protein expression and differentiation of tumor, in the manner of less differentiated tumors,show more P53 expression. Discussion: In regard to the absence of follow-up of the patients and informations about these protein and prognostic factors was performed indirect according to regional lymph node metastasis, tumor differentiation and tumor location. So, with follow-up the patients in prospective studies, the study of correlation between expression of a fore mentioned proteins with the survival is suggested, and in the next stage, after confirmation of correlation between expression of these proteins and survival, the measurement of these protein in serum as serum markers will continued. It is proposed to evaluate this correlation ina prospective study so that future investigations might lead to demonstration of a serum marker for early diagnosis of this lethal malignancy.
