Effects of Verapamile and Naloxone on Suppression of Seizures in Pentylenetetrazol-Kindled Rats

Background: Regarding the prevalence and complications of seizure attacks and necessity to control them, this study was carried out in order to assess the role of opioid receptors and voltage dependent calcium channels on the kindling induced by the convulsant pentylenetetrazol (PTZ). Materials and Methods: This experimental study was carried out on 24 wistar rats. In order to induce chemical kindling, PTZ was injected in sub-convulsive doses (37.5 mg/kg i.p) every 48 hours and effects were monitored for. 20 minutes and convulsive responses classified on the basis of 5 point-scoring system. Verapamile (calcium channel blacker), naloxone (opioid antagonist) or both and naloxone-verapamile solvent were administrated 20 min before PTZ application. The effect of these chemicals on seizure stage, period of stage 5 and reverse period of entering stage 5 was observed. The data were analyzed through Wilcoxon statistical test. Results: Verapamile significantly reduced the period of stage 5 and reverse period of entering stage 5. Naloxone had no significant effect on seizure expression of fully kindled animals, whereas application of naloxone and verapamile together had profound preventive effects on all seizure parameters. Conclusion: The results of this study suggest that naloxone and verapamile have synergistic suppressive effect on seizure induced by PTZ kindling. Further studies in this field are recommended.
