The Isfahan isolate of Eggplant mottled dwarf virus (EMDV) was transmitted mechanically to Nicotiana Tabacum var. Turkish and N. glutinosa. EMDV virions were purified by homogenization of infected N. tabacum var. Turkish and N. glutinosa tissue in citrate buffer and acidification of the extract to pH 5.4 followed by differential and step-gradient centrifugation. Purification of nucleocapsids was carried out by adding 2% Triton X-100 to the extract followed by differential and density gradient centrifugation. Both virion and nucleocapsid preparations were used to inject rabbits for antiserum preparation. Antiserum to EMDV virions produced two precipitin lines with homologous antigen while antiserum to EMDV nucleocapsids produced a single precipitin line with homologous antigen in agar gel diffusion test. In SDS- polyacrylamide gel electrophoresis, EMDV showed five protein bands with molecular weights of 82.5-83, 62-63, 57, 31-32 and 20-20.5 kDa. These values correspond to those of proteins G, N, NS, MI and M2 in rhabdoviruses, respectively. EMDV had a single nucleic acid species with a molecular weight of 4.028 * 10^6 in agarose gel electrophoresis.
