According to reports of ornamental and flowering office_ deputy of horticulture Ministry of Jihad-E-Agricultureʹ the area under ornamental cultivation in Iran, in 1996 was 3460 hectars of which 24% were in Mahallat area In respect to the importance of ornamental plants, Ornamental crops including: Tropaeo majus, Rosa spp., Gaillardia grandiflora,
Calandula spp., Gompherena spp., Petunia hybrida, Oenothera biennis, Tagetis spp., Althea spp. , Dahlia spp., Dianthus spp., Chrysanthemnm spp., Gladiolus spp_, Rudbekia spp., and Anthirrhiraum spp. with or without any obvious virus like symptoms were collected from Mahallat (Markazi) and Varamin (Tehran) growing areas showing: spotted wilting, leaf necrosis, chlorotic and stunting. Sampling included cut or whole flowering plants. Symptomatic leaves or stems were collected and transferred to research glasshouse. Samples were tested by double (or triple TAS-ELISA according to each antibody description) antibody sandwich enzyme linked immunosurhent assay (DAS-ELISA) using polyclonal antibody against tomato Varamin virus (TOVV) (As No. 0526), TAS-ELISA with monoclonal antibody (As No. 0106/As-0105) against tomato spotted wilt virus (TSWV.) and Impatiens necrotic spot virus (INSV) (As No. 117/As-0115) gifted from DSMZ,Braunschwing. Germany, Individual thrips were separated from the flowers by a fine-tipped brush. 2-3 thrips were removed and transferred to each 1.5 ml test tubes (eppendorf), and 100 miul ELISA extraction buffer were added to each tube. The tubes then were incubated 24 h at 4 °C. The content of the test tubes were macerated with the blunt end small glass rod and the extract was transferred to ELISA plate wells, as for an antigen coating stage. Meanwhile Thrips species were also preserved in 70° ethanol for further studies. Identification of individual thrips species were on the basis of observation of microscopic slides prepared for each adult thrips. The ELISA results with individual thrips indicated Thrips tabaci Lind, and Microcephalothrips abdominalis which infested Althea spp., Oenothera biennis and T. tabacci From Mahallat Dahlia spp. (var. short stem) were positive to TSWV. T.tabaci. and Frankliniella tenuirnis Uzel. collected on Rosa spp, and T tabacci Collected from Tagetis spp. (Varamin area) were positive with antibody to TOVV. T.tabacci in ELISA test Chrysanthemum spp. from Mahallat was found to be doubly infected with TOVV and INSV. The thrips infested plant samples were also tested and found positive to the respective antibody in ELISA test. Insect transmission test and determination of Thrips sp. efficiency for transmitting each tospoviruses requires further investigations. The results can be applied to monitor viruliferous thrips population and forecasting disease epidemics, to device a strategy for a virus disease control management.
