Silymarin, the extract of milk tistle (silybiim marianum\ is a strong antioxidant and it scavengers free radicals. Some drugs may cause nephrotoxicity by oxidative reactions; for example, oxytetracycline may damage renal tubules in long-term or large dose use. In the present study the role of silymarin was investigated to prevent nephrotoxicity produced by oxytetracycline in rabbits.
In present study, in four groups of rabbits (each group 6 rabbits each group) the following experiment was conducted: Group 1: normal saline, group 2: oxytetracycline (15 mg/kg), group 3: oxytetracyclme (15 mg/kg) + vitamin E (25 mg/kg) and group 4: oxytetracyclme (15 mg/kg) + silymarin (200 mg/kg) were injected intramuscularly everyday for 40 days. Finally, blood samples were collected from the rabbits and the serum quantity of creatinin and BUN were measured through laboratory kits.
The mean of creatinin increased significantly in group 2 but not in group 1. This mean decreased significantly by silymarin and vitamin E in groups 3 and 4. The mean of BUN increased significantly in group 2 and not in group 1 and this decreased in groups 3 and 4. Thus, long-term administration of therapeutic dose of oxytetracycline can impair the function of kidney but silymarin and vitamin E have protective effect on kidney against this toxicity.
