چكيده لاتين :
Tissue plasminogen activator protein was found as superior therombolytic agent for treatment of diseases such as acute myocardial infarction and efforts is currently focused on improving t-PA molecule and thereby itʹs pharmacokinetic properties. Reteplase (K2S) is a derivative t-PA that has a longer half-life and greater resistance to inhibitor than the natural t-PA molecule. The aim of this research is cloning of K2S form of the t-PA cDNA in eukaryotic system L. tarentolae which is recently has been introduced as a suitable host for expressing eukaryotic genes. K2S form of t-PA cDNA amplified by PCR and has cloned at ssurRNA (18s) site of L. tarentolae genome by homologous recombination. Hence, L. tarentolae is the first useful biotechnologically eukaryotic organism and displays rapid growth rates and simple nutrient requirements; we hope to reduce the production cost of this protein by cloning K2S gene in this system.
