Molecular characterization of chitin synthase gene in dermatophyte pathogen Trichophyton verrucosum

Trichophyton verrucosum is one of the dermatophyte fungi which invades the skin, hair of human and animals especially cattle. Several properties of this fungus have been investigated so far. However a few studies were carried out in the field of molecular biology of this fungus. The main aim of this survey was identification of the chitin synthase gene which is responsible for synthesis of chitin in this fungus. Paris of 21 nucleotide primers were designed from highly conserved regions of the similar genes in other fungi. Mentioned primers were utilized in PCR by using isolated genomic DNA of Trichophyton verrucosum whereas the PCR fragments were then sequenced. Eventually, 603 nucleotides have been sequenced from this new gene which encodes a polypeptide whit 201 amino acids. Analysis of this protein indicates that it has large amount of glycine and lysine but it is poor in tryptophan and cysteine. Sequences comparison in gene data bank (NCBI, NIH) for this part of DNA and its deduced amino acid revealed significant homology with members of the eukaryotic chitin synthase.
