چكيده لاتين :
Background; Fasciolosis is a worldw ide disease with major economic and public health
consequences. Ea rly detection of the infec tion is important for the prevention and contro l
of the disease . ELISA allows for ea rly detect ion of fascio los is in man and anima ls.
Fasc iolosis is ca used by Fasciola hepatica and F. gigantica in man and domestic animals
respective ly. These two species have many similar morphological characterist ics. In th is
study, the crude an tige ns of these two species are inves tiga ted by ELISA test.
Methods: The excretory-secretory and somatic ant igens of two spec ies were prepared
from adult flukes collected from the bile ducts of shee p and stored at _20°(ʹ. For the
preparation of the antisera, the antigens we re injected to laboratory- bred rabbi ts. Each
rabbit rece ived five injections at interva ls of seven days, start ing wi th 0.5 ml and ending
with 2.5 m!. Ten days after the last injection, the rabbits were ble d, and serum samples
separated and stored at _20~c . The reaction between homologous and he tero logous
antigens and antise ra was tested by ELISA and opt ica l densities we re recorded.
Results; Excretory - secretory and somatic antigens of eac h species showed a strong
positi ve reaction with the an tisera of the other species. In a homologous combinat ion of
an tigens and ant isera, a stronge r reaction was observed compared to the het erologous
combina tion, therefore many antigen ic materia ls of both species are the same.
Conc lus ion : The differences of these crude antigenic mater ials of F. hepatica and F.
gigantica are ins ufficient to preven t cross reaction of two species by ELISA. Furt her
inves tiga tions are recommended for the identification, detection and purification of
antigenic materia l of each species to improve the specific ity of this assay
