Induction and optimization of callus initiation in sugarbeet {Beta vulgaris L.)

In order to determine hormonal composition and plant tissue type for callusing optimization, three factor including (monogerm and multigerm), explant (hypocotyl, cotyledon) and hormonal composition (1 mg/1 benzyl adenine or 1 mg/1 2,4-D+ 0.5 mg/1 kinetin) were investigated as a factorial experiment with completely randomized design in 8 treatments and 4 replications. The explants were placed on solid MS medium containing the hormonal components. After one month, characters such as callusing percent and callus growth were measured. The results of analysis of variance showed that the highest callusing percent and callus growth is obtained from hypocotyl of multigerm line in the medium contained 1 mg/1 2,4-D and 0.5 mg/1 kinetin. With respect to friability of hypocotyl-derived callus in the medium contains 2,4-D and kinetin, it was concluded that this treatment can be used to prepare cell suspension culture in the future studies.
