A survey of contamination rate on Lahijan surface and sub-surface waters to Giardia lamblia by Real-Time PCR

Background and Aim: Giardia in one of the most common human parasite that is routinely transmitted via consumption of water to determine the Giardia-contaminated water resources of Lahijan surface and sub-surface waters in terms of parasitic contamination, the rate of Giardia DNA isolation was assayed by Real-Time PCR method. Materials and Method: During a cross-sectional study 169 water samples were collected from many different locations from water resources in Lahijan and outskirt of city and checked. The water samples were first filtered through (3 Mm pore size) the filter papers were washed with distilled water and the collected water was centrifuged. The remaining pellets were subjected to DNA extraction followed by regular PCR with G.lamblia specific primers. PCR positive samples were subjected to Real-Time PCR for further conformation. Results: Only 6 out of 169 (3.5%) water samples were appeared to be contaminated with Giardia positive that were collected during the autumn and winter in year 1385. Out of those positive samples, 4 were collected from river waters, I was from an east city residential well water, and the remaining 1 sample was from man-made waterfall. All of these positive samples were confirmed by Real-Time PCR. Conclusion: In reference to the fact that almost all positive samples were close to a sewage or slow-flowing river source, it reveals a strong correlation between the two. However, a more comprehensive study is required in order to fully evaluate the exact source of Giardia contamination.
