Study of Damask rose genetic diversity by RAPD and morphological markers

Rosa damascena Mill., an important species as a source of rose oil, is one of the oldest rose species grown in Iran. However genetic improvement of this species has been limited because of less information on its genetic variability. Therefore, an investigation was conducted at faculty of Agriculture of Tehran University. The genetic relationships among 41 Damask rose accessions from different areas of Iran (Fars, Isfahan, East Azerbaijan, Kerman and Razavi Khorasan provinces) and one accession from Bulgaria were studied using 31 RAPD (Randomly Amplified Polymorphic DNA) primers and 10 morphological markers. Both RAPD and morphological markers revealed high level of polymorphism among studied rose genotypes. Genotypes were divided into 10 groups based on RAPD marker comparing to 3 for morphological markers. The degree of discrimination was much less for the morphological data comparing to RAPD marker that was revealed by less numbers of separated groups. This study showed that studied Damask rose genotypes has aconsiderable genetic variation and potentially represents a rich and valuable gene pool of this species for breeding programs in future.
