Micropropagation of Mature Trees of Eucalyptus microcarpa and E. camaldulensis

Eucalyptus genus belongs to Myrtaceae family, which are exotic in Iran. Asexual propagations of E. camaldulensis Dehnh. and E. Microcarpa Miaden through stem cutting and grafting are very difficult. Therefore, micropropagation of mature trees developed in this research, using regenerated shoots from auxiliary bud explants. The trees were selected on the basis of their better growth rate, physical and phonotypical characteristics and free from diseases. Micropropagation was obtained on modified MS medium. The best sterilization treatment was obtained from 0.1% (w/v) HgCh solution for 4 minutes and then three times washing with sterile double-distilled water. Subcultures were made each six weeks. The most plantlets were obtained from explants which had been harvested on Oct-Nov. The best shoot formation occurred in both species on the medium containing 0.01 mg/1 TDZ and 0.5 mg/1 NAA. Also well rooted shoots produced in hormone-free MS medium. The plantlets were gradually acclimatized to ex vitro condition and transferred to soil successfully.
