مطالعه آسيب‌شناسي آرتريت ناشي از كلاژن تيپ2 گاوي و ‌اَجوان كامل فروند دررت

PATHOLOGICAL STUDY OF ARTHRITIS INDUCED BY TYPE 2 BOVINE COLLAGEN AND FREUNDʹS COMPLETE ADJUVANT IN RAT

به‌د‌ليل شباهت‌هاي آسيب شناختي بين آرتريت روماتوييد انسان با آرتريت ناشي از تزريق كلاژن در جوندگان، پژوهش هاي زيادي در اين زمينه صورت گرفته است. هدف از مطالعه حاضر بررسي ضايعات آسيب‌شناختي آرتريت تجربي ناشي از كلاژن بود كه بر روي 50 سر رت ماده از نژاد Rattus norvegicus صورت گرفت. آرتريت با تزريق داخل جلدي (2 تزريق به فاصله 14 روز) محلول كلاژن تيپ 2 گاوي به همراه ياور كامل فروند در قاعده دم و كف پاي راست در 40 رت ايجاد گرديد. ابتلاي اندام حركتي با 4 درجه خفيف، ملايم، شديد و خيلي شديد نشان داده شد. در ارزيابي باليني، تعداد 22، 12، 5 و 1 رت به درجه خفيف، ملايم، شديد و خيلي شديد مبتلا شدند. تزريق در قاعده دم با شدت كمتر ضايعات همراه بود. مفاصل مبتلا بزرگ، متورم و سفت و در موارد شديد آنكيلوز و دفورميتي آنها جلب توجه مي‌كرد. زخم‌هاي عميق در كف پنجه پا در 14 مورد قابل تشخيص بود. درمطالعه ريزبيني، نفوذ سلول‌هاي تك هسته‌اي و هيپرپلازي پاپيلر لايه سينوويال غشاي مفصلي در موارد خفيف و ملايم، رشد بافت فيبروواسكولار (پانوس) برروي غضروف مفصلي همراه با تخريب آن در موارد شديد و تخريب كامل غضروف مفصلي و دسترسي بافت جوانه اي به استخوان ساب كندرال در مواد خيلي شديد مشاهده شد. در اين مطالعه ضايعات مفاصل مبتلا، با ضايعات بيماران مبتلا به آرتريت روماتوييد، پلي آرتريت اروزيو (تخريشي) سگ‌ها و آرتريت™آنسفاليت بزان همخواني دارد. ‌ ‌ ‌

Because of the pathological similarities between collagen induced arthritis (CIA) and rheumatoid arthritis, the CIA model has been the subject of extensive investigations. This study initiated to evaluate the pathologic lesions of induced arthritis by collagen in a total of 50 female rats (Rattus norvegicus.). Arthritis was induced by two intradermal injections (14 days interval) of type 2 bovine collagen solution and Freundʹs complete adjuvant (FCA) into the tail and right leg paw of 40 rats. Severity of arthritis in affected leg was represented in grade 1 to 4. Three weeks after beginning of the experiment, 22,12,5 and 1 affected rats were clinically diagnosed with 1, 2, 3 and 4 grades of arthritis respectively. Rats were intradermally injected into the tail showed only slight and moderate (grade 1 and 2) arthritis. Macroscopically, the involved joints were enlarged, swollen and stiff and deformity and ankylosis of affected joints were noticed in severe cases. Deep healing ulcers in planter of the paw were diagnosed in 14 rats. Histopathologic examinations revealed infiltration of mononuclear inflammatory cells and papillary hyperplasia of synovial membrane in mild and moderately affected animals (grade 1 and 2) and erosion of articular cartilage with formation of fibrovascular pannus on its surface in synovial membrane in severe cases (grade 3). In very severe cases (grade 4), complete destruction of articular cartilage and access of granulation tissue to subchondral bone were noticed. Pathologic lesions of the involved joints were very similar to those reported from rheumatoid arthritis in human, erosive polyarthritis in dog and caprine arthritis/encephalitis.
