Adjuvant properties of Brucella abortus OPS conjugate to TT for induction of antitetanus antibodies in versus purified tetanus toxoid

Tetanospasmin is a neurotoxin that causes tetanus. Recently the immunogenic and adjuvant properties of the O-chain (OPS) of B.abortus were under consideration. On the other hand, the OPS of B.abortus showʹs various adjuvant activities and improve immunological responses. This study aimed to investigate the adjuvant properties of OPS-TT conjugate against tetanus. OPS was extracted with 2% acetic acid solution and purified using Sephadex G-50 medium. Conjugate vaccine was prepared by ammida-tion method. Then three groups of female BALB/c mice (each group included 15 mice) were selected. Vaccination was performed by three doses with two weeks interval. Then serum samples were collected and antibody responses against tetanus were measured by ELISA method for total IgG, IgG1. IgG2a, IgG2b, IgG3, IgM, and IgA. Two weeks after first dose, there was no significance different between groups that were immuni-zided by TT and OPS-TT. But after second and third doses. OPS-TT showed signifi-cante increasing in all types of antibodies concentration against tetanus in versus TT. The anti TT IgG antibody was predominant in IgGl subtype and then IgG3, IgG2a, and IgG2b. These results show that OPS from B.abortus increases antitetanus antibodies in conjugate form with tetanus toxoid and can be an appropriate and effective adjuvant.
