Cytotoxic Effects of I.V. Injection of Urografin 76% on Spermatogenesis in Animal Model

Nowadays ionic contrast materials are being used in various procedures of radiological examinations all around the world. There are not many researches on cytotoxic effects of these materials on spermatogenesis. In this study we have investigated the effects of urografin 76% (an ionic iodinated contrast material) on spermatogenesis in mouse model. The contrast material was injected (2.5 ml/kg) to 5 groups of 3 male Swiss albino mice. Each group of mice was killed in the 18, 20, 22, 24, and 26 days after injection respectively and the sperm heads were counted and the maximum effect of the contrast material on spermatogenesis was seen in 20 days after injection. Then different doses of 0.5, 1, 2, 3, 4 ml/kg of urografin 76% were injected via tail vein of the 5 groups of 5 mice. The mice were killed 20 days after injection and their testicular sperm heads were counted using haemocytometer under a light microscope (x400). The mean numbers of sperm heads in injected groups in comparison with control group showed statistically significant differences in all doses of urografin 76% (p<0.001). The most decrease in the number of sperm heads occurred at lowest dose (0.5 ml/kg). It is recommended that using of urografin 76% to be restricted in patients with known history of fertility problems.
