مقايسه برون تني اثرات ضدميكروبي و سميت سلولي دهان‌شويه ليسترين و ايرشا

In vitro Comparison of antimicrobial activity and cytotoxic effects of Listerine and Irsha mouthwashes

چكيده مقدمه: دهان‌شويه ايرشا نمونه داخلي دهان‌شويه ضد باكتري ليسترين است. با وجود اين، تاكنون اثرات بيولوژيك آن، چه از نظر باكتريولوژي و چه از نظر سميت سلولي، با دهان‌شويه ليسترين كه اثرات مفيد اثبات شده دارد، مقايسه نگرديده است. هدف از اين تحقيق مقايسه اثرات ضدميكروبي و سميت سلولي دهان‌شويه ليسترين و ايرشا بود. مواد و روش‌ها: در اين مطالعه تجربي برون‌تني، اثر ضدميكروبي 5 رقت دهان‌شويه ايرشا و ليسترين (25/6، 5/12، 25، 50 و 100 درصد)، در زمان‌هاي مختلف بر روي غلظت 5/0 مك فارلند 5 سوش باكتري (استرپتوكوك موتانس، استرپتوكوك سانگوييس، انتروكوك فكاليس، كانديدا البيكانس و اشرشياكلي) ارزيابي گرديد. آزمايش براي هر باكتري 3 بار تكرار شد و از آب مقطر به عنوان كنترل استفاده گرديد. براي بررسي اثر سميت سلولي، بيش از 100000 سلول از رده‌هاي KB (اپي‌تليال)،MRF (فيبروبلاست)، J774.A1 (ماكروفاژ) و Saos-2 (استيوبلاست) در پليت‌هاي 96 خانه‌اي كشت داده و به مدت 72 ساعت در مجاورت 4 رقت از دو دهان‌شويه (5/0، 1، 5 و10 درصد) انكوبه شد. سپس سلول‌ها شسته و به مدت 45 دقيقه با MTT (Cloorimetric assays) رنگ شده و در نهايت نتايج با دستگاه اليزا ثبت شد. سپس نتايج در 18SPSS با آزمون‌هاي آناليز واريانس Tukey‌ و One Way ANOVA تفسير شد (05/0 = ?). يافته‌ها: اثر سميت سلولي دهان‌شويه‌هاي ليسترين و ايرشا در غلظت‌هاي مختلف از لحاظ آماري تفاوت معني‌داري نداشت (05/0 < p value). از لحاظ فعاليت ضد ميكروبي، دهان‌شويه ليسترين در دو رقت 100 و 50 درصد مانع از رشد تمام گونه‌هاي باكتريايي شد؛ در حالي كه در رقت‌هاي مشابه از دهان‌شويه ايرشا تنها 3 نوع باكتري رشد نكرد. نتيجه‌‌گيري: با توجه به نتايج به دست آمده، احتمال مي‌رود اثر ضدميكروبي دهان‌شويه ليسترين نسبت به ايرشا بهتر باشد، اما از لحاظ سميت سلولي تفاوت معني‌داري بين اين دو وجود ندارد. كليد واژه‌ها: ليسترين، ايرشا، دهان‌شويه، ضد ميكروبي، سميت سلولي.

Abstract Introduction: Irsha is an Iranian sample of Listerine mouthwash. To date biologic effects of Irsha and Listerine have not been compared. This study compared the antimicrobial and cytotoxic effects of Listerine and Irsha mouthwashes. Materials and Methods: In this in vitro study, antimicrobial effects of 5 dilutions of Irsha and Listerine, on the 0.5 MacFarlnd concentrations of five bacterial strains (Streptococcus mutans, Streptococcus Sangius, Enterococcus Fecalis, Candida albicans and Escherichia coli) were assessed at different times. Each test was repeated three times, with distilled water as control. For cytotoxic effect, more than 100000 cells of KB (epithelial cell), J774.A1 (macrophage), Saos-2 (osteoblast) and MRF (fibroblast) were cultured and incubated for 72 hours in different dilutions of the two mouthwashes (0.5, 1, 5, 10%). After incubation, the cells were washed and stained with MTT for 45 minutes. The results were recorded with ELISA READER and analyzed with SPSS 18 software, using ANOVA, Tukey test and one-way ANOVA. Statistical significance was defined at p value < 0.05. Results: The cytotoxic effects of various dilutions of Listerine and Irsha mouthwashes were not significantly different (p value > 0.05). In terms of antimicrobial activity, in dilutions of 100% and 50% of Listerine none of the five bacterial strains grew. However, at the same dilutions (100% and 50%) of Irsha, only three strains did not grow. Conclusion: According to the results, antimicrobial activity of Listerine is probably better than Irsha, but cytotoxic effects of Listerine and Irsha mouthwashes were not different.
