Evaluation of Triticum dicoccoides populations for stripe rust resistance gene Yr15 using locus specific marker SC792

Stripe rust is one of the most important fungal diseases of wheat, reported in more than 60 countries in the world. Wild emmer wheat (Triticum dicoccoides), the tetraploid progenitor of cultivated wheat, has proven to be a valuable source of novel stripe rust resistance genes for wheat breeding. G25 accession of this species has Yr15 gene conferring resistance to 31 stripe rust isolates. This gene cosegrageted with the codominant PCR-based locus specific marker SC792 on chromosome 1BS. This marker was used to screen T dicoccides genotypes carrying Yr15. Forty-four individuals out of 149 accessions from 16 populations were shown to amplify resistant allele of SC792. Genotypes of populations Yehudiyya and Tabigha1 didnʹt amplify any product for this marker. Results showed five alleles for marker SC792 in the studied populations and the marker was confirmed as a multi-allelic marker. Based on pathological test, all the individuals from populations Hermon and Roshpinna which produced an infection type similar to that of G25, amplified resistant allele of SC792, confirming the presence of this gene. Results demonstrated that marker SC792 can be applied to pyramidise Yr15 with other resistant genes to produce strip rust resistant varieties in wheat breeding programs.
