Evaluation of amplification quality of 17 bovine Y-chromosome specific microsatellites in sheep

In this study amplification quality of 17 bovine Y-chromosome specific microsatellites was evaluated in five Iranian native sheep breeds including Grey-Shiraz, Torki-Ghashghai, Afshari, Farahani and Naini. Thirty males and two females of unrelated sheep were randomly selected from each breed. Genomic DNA was extracted from blood using salting out method. Three loci (UMN0301, UMN2706 and UMN0803) produced smear bands and three (UMN0108, UMN0307 and UMN2405) presented ladder-like bands. Due to the fact that microsatellite locus in a Y-specific chromosome region can include multiple copies distributed along the chromosome, the observed smear and/or ladder like bands in PCR products obtained from these Y-chromosome microsatellite markers can be indication of existence a large copy number of these markers on Y-chromosome. Four loci (UMN3008, UMN0406, UMN0504 and UMN2713) were monomorphic. The remaining seven loci (UMN2303, UMN092, UMN0907, INRA124, INRA189, INRA057 and BM861) were found to be polymorphic. In total, 71 alleles were observed in these seven microsatellites, the highest and the lowest number of alleles were observed in INRA124 (17 alleles) and UMN2303 (4 alleles), respectively. The results of this research indicated that bovine Y-chromosome specific microsatellites can be used for determining genetic diversity in sheep.
