تحليل ارتباط ژنتيكي برخي هاپلوتيپ‌ها با ميزان كلسترول ـ HDL، تري‌گليسيريد و دور كمر به كمك روش Haplotype Base Association Test، در خانواده‌هاي داراي افراد مبتلا به سندرم متابوليك

Genetic Association of Some Haplotypes to Level of HDL-C, Triglyceride, and Waist in Family Members with Metabolic Syndrome Using Haplotype Based Association Test

مقدمه: هاپلوتيپ‌ها در بررسي ارتباط ژنتيكي، از اهميت ويژه‌اي برخوردار هستند. يكي از روش‌هاي بررسي ارتباط ژنتيكي هاپلوتيپ‌ها با يك يا چند صفت، استفاده از روش Haplotype Base Association Test (HBAT) است. آماره‌ي آزمون اين روش كه براي هريك از هاپلوتيپ‌ها محاسبه مي‌شود، از توزيع نرمال استاندارد تبعيت مي‌نمايد. در پژوهش حاضر به كمك روش HBAT، به بررسي ارتباط ژنتيكي هاپلوتيپ‌هاي برخي ميكروستلايت‌هاي منتخب با ميزان كلسترول ـ HDL، تري‌گليسيريد و دور كمر براي شناخت نواحي ژني موثر در سندرم متابوليك پرداخته شد. مواد و روش‌ها: تعداد 125 خانواده از بين شركت‌كنندگان در مطالعه‌ي قند و ليپيد تهران به گونه‌اي انتخاب شدند كه دست كم يك نفر از اعضاي آن‌ها مبتلا به سندرم متابوليك (براساس معيار ATP III)، و دست كم دو نفر دچار كاهش ميزان كلسترول ـ HDL باشند. بررسي ارتباط ژنتيكي ميزان كلسترول ـ HDL، تري‌گليسيريد و دور كمر با هاپلوتيپ‌هاي به دست آمده از برخي ميكروستلايت‌هاي كروموزوم‌هاي 8، 11، 12 و 16، و با به كار‌گيري روش HBAT بررسي گرديد. يافته‌ها: 125 خانواده شامل 563 نفر (269 مرد و 294 زن) بودند. در بررسي ارتباط ژنتيكي از كروموزوم 8، ارتباط هاپلوتيپ‌ 2-2-2-2 با ميزان كلسترول ـ HDL و تري‌گليسيريد و از كروموزوم 12، هاپلوتيپ‌هاي 1-2-2 و 2-2-2 با ميزان تري‌گليسيريد و 2-1-1 با ميزان دور كمر معني‌دار شد (05/0P < ). نتيجه‌گيري: هاپلوتيپ‌ها، داده‌هاي كامل‌تري را براي بررسي ارتباط ژنتيكي فراهم خواهد آورد. اميد است با يافتن هاپلوتيپ‌هاي موثر بر ميزان كلسترول ـ HDL، تري‌گليسيريد و دور كمر توسط روش HBAT، راه‌كار مناسبي براي طراحي بررسي‌هاي بعدي به منظور يافتن ژن‌هاي مستعد كننده سندرم متابوليك فراهم آيد.

Introduction: Haploytpes are important elements in study of genetic associations. The haplotype based association test (HBAT) is a method to study genetic association of haplotypes with one or more traits. The test statistic in this method, which is calculated for all haplotypes, follows a standard normal distribution. In this study, in order to find the chromosomal area locus of genes affecting metabolic syndromes, the HBAT method was used to investigate the genetic association of haplotypes of some candidate microsatellites with HDL-C, triglycerides, and waist. Materials and Methods: A sum of 125 families with at least one member having metabolic syndrome according to ATPIII, and at least two members with low HDL-C levels were selected from among participants of the Tehran Lipid and Glucose Study (TLGS). The genetic association of HDL-C, triglycerides, and waist with haplotypes of some microsatellites of chromosome 8, 11, 12, and 16 was studied, using HBAT. Results: Data was obtained for 125 families, consisting of 563 individuals, aged 20 years or above (269 males and 294 females). Genetic association of the haplotype 2-2-2-2 of chromosome 8 showed significant association with HDL-C and triglycerides. Haplotypes 2-2-1 and 2-2-2 of chromosome 12 showed significant association with triglycerides. In addition, haplotype 1-1-2 of this chromosome was found to be associated with waist (P < 0.05). Conclusion: Researching haplotypes provides more information on genetic associations, and identification of haplotypes influencing HDL-C level, triglycerides, and waist may be helpful in designing future research aimed at determining the genes predisposing persons to metabolic syndrome.
