تغييرات هيستومرفولوژيك غده تيروييد موش سوري متعاقب تجويز طولاني مدت مرفين

Histomorphological changes of surri mouse thyroid gland following long term administration of morphine

زمينه و هدف: گزارشات ضد و نقيضي در مورد اثرات مرفين بر غده تيروييد وجود دارد. اين مطالعه با هدف بررسي تاثير استفاده طولاني مدت مرفين بر وزن بدن، وزن غده تيروييد و تغييرات هيستومورفولوژيك غده تيروييد موش سوري در مراحل مختلف رشد طراحي و اجرا شده است. روش بررسي: در اين مطالعه تجربي 150 سر موش سوري ماده نژاد Balb/c با سن دو هفته، به صورت تصادفي به سه گروه مساوي و در مرحله بعد موش ها در هر گروه به چهار زير گروه تقسيم شدند. زير گروه اول (زير گروه شاهد 5 =n) فقط آب، زير گروه دوم (زير گروه شم 5=n) آب و mg/ml5 شكر، زير گروه سوم (زير گروه تجربي يك 20 =n) mg/ml04/0 مرفين و mg/ml5 شكر و زير گروه چهارم (زير گروه تجربي دو 20 =n) mg/ml 4/0 مرفين و mg/ml5 شكر به صورت خوراكي دريافت كردند. گروه اول و دوم 21 روز و گروه سوم 63 روز مرفين مصرف كردند. داده ها با استفاده از آزمون آماري ANOVA و تست تعقيبي Tukey در نرم افزار SPSS مورد تجزيه و تحليل آماري قرار گرفتند. يافته ها: مصرف مزمن مرفين در هر دو غلظت باعث كاهش چشمگير وزن بدن موش ها در زير گروه هاي دوم و سوم و همچنين وزن تيروييد در زير گروه هاي اول و دوم شد (05/0 > P). مطالعات هيستومرفولوژيك آشكار كرد كه مصرف مزمن مرفين باعث افزايش اندازه و شكل سلول هاي فوليكولي در تيروييد شده است؛ افزون بر اين شكل فوليكول هاي غده در زير گروه هاي تجربي تغيير كرده و حاوي مقادير كمي كلوييد در مقايسه با زير گروه هاي شم و شاهد شده است. نتيجه گيري: مصرف مزمن مرفين مي تواند علاوه بر وزن بدن و وزن تيروييد، ساختمان و فعاليت تيروييد را نيز تحت تاثير قرار دهد.

Background and aims: There are contradictory reports about the effects of morphine on thyroid glands. The aim of the present study was to investigate the effects of long term use of morphine on the body weight, thyroid glands’ weight and the histomorphological structure of mouse thyroid gland at different stage of growth. Methods: In this experimental study 150 two weeks female bulb/c mice were randomly divided into three equal groups. The mice in each group were divided into 4 subgroups, subsequently. Subgroup one (control group; n=5) accessed to clean water. Subgroup two (sham group; n=5) received 5mg/ml sugar, orally. 0.04mg/ml morphine and 5mg/ml sugar were available for subgroup three (experimental group one; n=20). 0.4mg/ml morphine and 5mg/ml sugar were given orally to subgroup four (experimental group two; n=20). The first and second group received morphine for 21 days and the third group received morphine for 63 days. Results of investigating the mice and their thyroid glands weight were analyzed using ANOVA and Tukey tests, results of the histomorphological studies were reported as a descriptive report. Results: The results showed that chronic use of morphine with both doses significantly reduce body weight of mice in group two and group three (P < 0.05). Moreover, chronic usage of morphine with both doses significantly reduced thyroid weight in group one and two (P < 0.05). Histomorphological studies revealed that chronic usage of morphine increased the size and shape of follicular cells in thyroid gland. Furthermore, the shape of follicles of thyroid gland in experimental groups were changed and contained a little clouid compared to control and sham groups. Conclusion: Based on the results from present study it can be concluded that the chronic usage of morphine may in addition of its effect on the body weight and thyroid gland weight have influence on the structure and function of thyroid gland.
