پيوستگي پلي مورفيسم (C/T593+) ژن GPx1 با بيماري آرتريت روماتوييد در استان گيلان

Association of Glutathione peroxidase 1(+593C/T) gene polymorphism with rheumatoid arthritis in Guilan province

چكيده زمينه و هدف: آرتريت روماتوييد، يك اختلال التهابي مزمن است كه اساسا روي مفاصل محيطي و بافت‌هاي اطراف آن اثر مي‌گذارد. فاكتورهاي ژنتيكي و محيطي در پاتوژنز اين بيماري موثر مي‌باشند. در سال‌هاي اخير، نقش استرس اكسيداتيو و آنزيم‌هاي دخيل در اين فرآيند، در آرتريت روماتوييد مورد توجه قرار گرفته است. در مقابله با گونه‌هاي واكنش‌پذير اكسيژن، آنزيم‌هاي آنتي‌اكسيداني دروني و گلوتاتيون پراكسيدازها نقش برجسته‌اي بازي مي‌كنند. از پلي مورفيسم‌هاي مهم اين ژن، تغيير كدون CTC به جاي كدون CCC بوده كه موجب جانشيني اسيدآمينه لوسين در موقعيت 198به جاي اسيدآمينه پرولين مي‌شود. هدف از اين پژوهش، بررسي ارتباط پلي‌مورفيسم ژن GPx1 در بيماران مبتلا به آرتريت روماتوييد مي‌باشد. مواد و روش‌ها: در اين مطالعه مورد- شاهدي 130 بيمار و 126 فرد سالم مورد بررسي قرار گرفت.DNA ژنومي از سلول‌هاي خون استخراج شد. ژنوتيپ‌ها با استفاده از تكنيك PCR-RFLP تعيين شد. آناليز داده‌ها با نرم افزار MedCalc (ويرايش 1/12) صورت گرفت. يافته‌ها: فراواني ژنوتيپ‌هاي CC، CT و TT به ترتيب در بيماران 31/32، 08/43 و 61/24 درصد و در افراد سالم 62/42، 10/54 و 28/3 درصد محاسبه شد. نتايج آماري نشان دهنده ارتباط معني‌داري بين ژنوتيپ TT ژن GPx1 و بيماري آرتريت روماتوييد بود(002/0p=). نتيجه‌گيري: نتايج حاصل از اين پژوهش نشان‌دهنده نقش ژنوتيپ TT در ارتباط با بيماري آرتريت روماتوييد در افراد مورد بررسي مي‌باشد. هرچند جهت تاييد نتايج، پيشنهاد مي‌گردد كه مطالعه در جمعيت بزرگ‌تري صورت گيرد. واژگان كليدي: پلي مورفيسم ژني، GPx1، آرتريت روماتوييد

Abstract Background: Rheumatoid arthritis is a chronic inflammatory disorder that mainly affects the peripheral joints and surrounding tissue. Genetic and environmental factors have important roles in pathogenesis. The role of oxidative stress and the enzymes involved in the pathogenesis of rheumatoid arthritis have been studied in these years. A predominant role in counteracting reactive oxygen spieces is played by endogenous antioxidant enzymes, and glutathion peroxidase (GPx). An alteration of CTC codon to CCC codon which results in substitution of leucine instead of proline in 198th amino acid location is one of the important polymorphisms of this gene. The aim of this study was to evaluate the association of GPx1 gene polymorphism in patients with rheumatoid artheritis. Materials and Methods: The case-control study included 130 patients with rheumatoid artheritis and 126 healthy individuals. Genomic DNA was extracted from white blood cells and the genotypes were identified using polymerase chain reaction-restriction fragment length polymorphism (PCR - RFLP). Statistical analysis was carried using the MedCalc (version 12/1). Results: The frequencies of CC, CT and TT genotypes of GPx1gene were 32/31%, 43/08% and 24/61% respectively, in rheumatoid arthritis patients, whiles in healthy volunteers were 42/62%, 54/10% and 3/28%. Statistical results showed significant relationship between TT genotype of GPx1 gene and Rheumatoid arthritis (p=0/002, ?2 =11/715, OR: 9/90 95% CI (2/04 to 48/01)). Conclusion: In conclusion, these results indicate that TT genotype of GPx1 gene may be associated with the risk of rheumatoid artheritis in the studied population. However, further research is required to clarify the role of gene polymorphism in rheumatoid arthritis. Keywords: Gene Polymorphism, GPx1, Rheumatoid Artheritis
