The effect of 5-HT2 serotonergic antagonist ritanserin on scopolamine-induced amnesia was investigated using Morris water maze (MWM). Adult male Wistar rats were divided into eight groups and CAI region of the hippocampus was bilaterally cannulated. One week later, they received vehicles or drugs through cannulae each day. Animals were tested for four consecutive days (4 trials/day) in MWM during which the position of hidden platform was unchanged. On fifth day, the platform was elevated above the water surface in another position to evaluate the function of sensory, motor and motivational systems. The results showed a significant increase in escape latencies and traveled distances to find platform in scopolamine (2 140.5 µl/side) - treated group as compared to the vehicle (saline) and a significant decrease in these parameters in ritanserin (4 )40.5 µl/side) -treated group as compared to the vehicle (DMSO). Furthermore, in scopolamine+ritanserin-treated group, a significant decrease in escape latency and traveled distances was found as compared to the scopolamine-treated group.
These findings clearly imply that microinjection of ritanserin into CAI region of the hippocampus improves the scopolamine-induced amnesia in male rats.
