Angiogenesis is a complex process during which new blood vessels are generated from the pre-existing vessels. This phenomenon plays an important role in embryonic growth, tissue repair, pathological processes including tumor growth, and so forth. In this respect, the development of tumors is dependent upon new vessel formation to provide enough nutrient and oxygen.
Considering the involvement of many factors in angiogenesis, this study was conducted to evaluate the effect of anti-plasminogen monoclonal antibody MC2B8 on angiogenesis. For this purpose, an in vitro model of angiogenesis was designed using fibrin gel and its covering of endothelial cells of bone marrow capillaries as a three-dimensional structure for endothelial cell growth. Then, different titers of monoclonal antibody MC2B8 (30-480 gg/ml) was added to fibrin gel. The fractions were collected n, 3-5 days after culturing of endothelial cells, growth and migration of cells and ensuing capillary development was microscopically observed. The results showed that addition of MC2B8 delays clot lysis and inhibits angiogenesis.
These findings suggest that the inhibitory effect of MC2B8 on angiogenesis is possibly mediated through blockade or induction of conformational changes in plasminogen epitopes that are involved in angiogenesis and fibrinolysis.
