ارزيابي فعاليت آنتي‌اكسيداني و ضدباكتريايي عصاره متانولي سه گونه از جنس Centaurea L. (مركبان) از ايران

Antioxidant and antibacterial activities of the methanolic extracts of three Centaurea L. species (Astraceae) from Iran

جنس Centaurea L. با حدود 700 گونه در دنيا و 70 گونه در ايران، بزرگ‌ترين جنس از تيره كاسني (Asteraceae) است. گياهان اين جنس بدليل خواص دارويي خود، در طب سنتي برخي كشورها كاربرد دارند. در اين مطالعه سنجش محتواي فنلي، فلاوونوئيدي، پتانسيل آنتي‌اكسيداني و خواص ضد‌باكتريايي عصاره‌ متانولي سه گونه‌ از جنس Centaurea شامل C. imperialis، C. iransharii و C. glastifoliaبه تفكيك در دو بخش كاپيتول و اندام هوايي (برگ و ساقه) انجام شد. سنجش محتواي فنل كل به روش فولين سيوكالتئو و سنجش محتواي فلاوونوئيد كل به روش كلريدآلومينيوم و ارزيابي خاصيت آنتي‌اكسيداني عصاره‌ها به روش مهاركنندگي راديكال آزاد DPPH انجام شد. همچنين خاصيت ضدباكتريايي عصاره‌ها به روش انتشار ديسك ارزيابي شد. محتواي فنلي عصاره‌ها قابل توجه‌ و بين 92/35 تا 73 ميلي‌گرم گاليك اسيد بر گرم عصاره و محتواي فلاوونوئيدي عصاره‌ها بين 45/4 تا 31/5 ميلي‌گرم كوئرستين بر گرم عصاره بود. تمام عصاره‌ها خاصيت مهاركنندگي بالاي راديكال آزاد نشان دادند و در بين آن‌ها، اندام‌هاي هوايي C. iransharii كمترين IC50 را به خود اختصاص داد (11/0= IC50 ميلي‌گرم بر ليتر). همچنين عصاره‌ها خاصيت ضد‌ميكروبي قوي در برابر باكتري‌هاي گرم مثبت Bacillus cereus ، Staphylococcus aureus ، Bacillus megaterium و Bacillus thuringiensis نشان دادند.

Centaurea L. with about 700 species in the world and 70 species in Iran is the largest genus in the family Asteraceae. Centaurea species are used for the treatment of various ailments in the popular medicine in some countries. The aim of the present study was to investigate total phenol and flavonoid contents, antioxidant potency and antibacterial activities of the methanolic extracts from capitulum and aerial parts of three Centaurea species, namely C. imperialis, C. iransharii and C. glastifolia. Total phenolic and flavonoid contents were evaluated by FolinCiocalteu and aluminum chloride methods, respectively. Antioxidant activity was evaluated by DPPH radical scavenging assay. In addition, antibacterial activities of the extracts were studied by disc diffusion method. Results from the study showed that total phenol content in the extracts were found to a range between 35/92 and 73 mg GAE/g extract and total flavonoid content varied from 4.45 to 5.30 mg QE/g extract. A significant antioxidant capacity was found in all studied extracts and aerial part extract of C. iransharii showed the highest antioxidant activity (IC50 = 0.11 mg/ml). In addition, the studied species represented strong antimicrobial activity against Gram positive bacteria.
