Effect of Serum on In Vitro Maturation of Immature Bovine Oocytes and Effect of Heparin and Semen From Different bulls on Embryo Cleavage Rate

The need for reproduction of bovine blastocysts from in vitro matured, fertilized and in vitro developed embryos for research and practical application is well known evident. It is necessary to adjust and optimize the in vitro environment so that the quality of developing embryos approaches the quality of in viva developing embryos. For this purpose 1225 immature Bovine oocytes were obtained from 2-8mm follicles. To study the effective factors on in vitro Oocyte maturation and fertilization and subsequent development is of great importance. This study evaluated use of serum from in vitro maturation of oocytes and the effect of different doses of heparin and sperm from different bulls on fertilization, cleavage rate as an index for in vitro maturation and fertilization rate. Sperms from different bulls were used to assess maturation, development and cleavage rate of 1225 immature bovine oocytes obtained from follicles to 2-8mm. and data obtained showed no significant difference between the different kinds of serum used as a protein supplementation for in vitro maturation. Iranian Heparin with doses of twenty units per/ml can enhance capacitation of spermatozoa and optimize cleavage rate (P=0.0068). The cleavage rate when using semen from two different bull were also significantly different (P=0.017). It can be concluded that for in vitro production of embryos from immature oocytes it is necessary to obtain the optimal does of heparin and semen from a suitable bull for optimal production of high quality embryos.
