Embryonic stem (ES) cells are pluripotent cells derived from the inner cell mass of blastocysts. Recently, the development of appropriate culture conditions for differentiation of these cells into specific types have permitted their use as potential therapeutic agents for several diseases. In addition, manipulation of their genome in vitro allows the creation of animal models of human genetic diseases and the study of gene function in vivo. This study started to produce new ES cell lines for future aforementioned studies. We used 3.5 day blastocysts recovered from C57BL/6 mouse strain and cultured on mouse embryonic fibroblasts in ES media 1000iu/ml and/or 5000iu/ml leukemia inhibitory factor (LW). Established lines were analyzed by simple karyotype, C-banding, SRY-PCR, alkaline phosphatase, and Oct-4 expression (RT-PCR). Two ES cell lines from C57BL/6 mouse strains were produced, One ES cell line established 1000 and 5000iu/ml LIF. Both ES cell lines had a normal karyotype. C-banding and SRY-PCR showed that all lines had a XY sex chromosome composition. Also, all lines had alkaline phosphatase activity and expressed Oct-4. These results indicate that two male murine ES cell lines with normal karyotype, alkaline phosphatase and Oct-4 positive properties were established from C57BL/6 strain.
