Abstract
: Chickpea (Cicer arientinum L.) is an important legume crop in Iran. Thus, an experiment was conducted to determine the leaf and seed storage protein profiles of four chickpea cultivars (under drought stress and non stress conditions) include Arman, Hashem, Azad and ILC-482 were analysis based on biochemical markers, by Sodium Dodecyl Sulfate Polyacrylamide Gel electrophoresis (SDS-PAGE). Leaves and seed proteins were purified by extraction buffer (Tris-HCL, 50Mm, pH 8.5 containing NP-40, 0.2%, PMSF, 1mM and EDTA, 1mM, MgCl2, 20 Mm and 2-mercaptoethanol 2%) and resolved in 12.5% and 5% resolving and stacking gels respectively. The results of SDS-PAGE for Leaves and seed storage proteins were showed 26 and 25 bands respectively. Seed protein most different related to Azad and ILC-482 cultivars. Based on analysis of cluster, in both conditions, genotypes Azad and ILC482 had the highest genetic distance and can be applied as parents in hybridization in plant breeding to get highest hybrid vigor. Ii is concluded that leaves and seed storage protein profiles could be useful markers in the studies of genetic diversity.
