Abstract :
ABSTRACT:Mistletoe (Viscum album) is a hemi-parasiticshrub which grows on the stems of other treesand absorb nutrients of them. It is used widely in medicine. In this study for the first time in Iran, the Randomly Amplified Polymorphic DNA (RAPD) technique was employed to determine genetic diversity of 17 genotypes of Mistletoe from Golestan, Gilan and Mazandaran provinces. Thirteenoligonucleotide primers were used to amplify the genomic DNA isolated from leaves. 13RAPD primers amplified 169 bands that 83.38 % bands were polymorphic. Average Polymorphism Information Content (PIC) was 0.27 and average Marker Index (MI) value was 20.7for RAPD markers respectively. Cluster analysis based on DICE coefficient and UPGMA algorithm showed that there is a wide variation in the collected samples. The average of similarity based on RAPD markers was 0.45. This high level of diversity could be related to the existents of several sub species in Mistletoe (Viscum album) and the high discrimination power of molecular markers. RAPD molecular markermay serve as a complementary tool to determine the genetic diversity.
