Dendritic Cells Bearing HLA-G Inhibit T-Cell Activation in Type 1 Diabetes

HLA-G is normally expressed o n hum an troph oblast cells. It is a no n-class ical MHC
molecule class I b. Th e role o f HLA -G in diab etic type 1 is not known.
We investigated th e role o f IFN-(BETA) in induction HLA-G expression o n th e monocyte
derived dendritic cells (DC) in diabetes type 1.
T reatm ent of dendritic cell with IfN-(BETA) in vitro from diabetic patients (n=20) and normal
subjects (n=20) resulted to the production and exp ression of HLA-G o n th ese cells from both
grou ps. However, comparison of DC from the diabetic patients with DC from the co ntrols
revealed lower levels o f HLA-G molecul es in DC from diabetic patients. Using mixed
lymphocyte reaction (MLR), it was found th at DC exp ressing f-1LA-G mediated the inhibition
of auto logo us T cell activatio n.
It is co ncluded that I FN - (BETA) can increase HLJ\-G in DC from diabetic pati ents; subse quently
it may prevent the immune regularly pathway in th e diabetic pathogenesi s
