Synthesis of a novel pyrimido[4,5-e][1,2,4]triazolo[3,4-b][1,3,4]thiadiazepine as a Novel Heterocyclic System

Pyrimidine and related fused heterocyclic compounds have attracted the attention of numerous researchers due to their important biological. they have been reported asanticancer, antiviral, antitumor, and anti-inflammatory agents [1-2].Moreover, tetrazoles have found utility in pharmaceuticals as lipophilic spacers and carboxylic acid surrogates, in special explosives, photography and information recording systems[3]. Herein, 2,4-dichloro- 5(chloromethyl)-6-methylpyrimidine 1were prepared according to the previous procedure and treated with sodium 1-amino-1H-tetrazole-5-thiolate 2 as abinucleophile in CHCl3. to obtain 5-((2,4-dichloro- 6-methylbenzyl)thio)-1H-tetrazol-1-amine3. Compound 3was further cyclized with K2CO3 in boiling DMF to give the desired novel tricyclic compound4. Treatment of compound 4 with various secondary amines afforded the corresponding products5a-gin yields(Figure 1). All of these compounds were characterized by FT-IR, 1H NMR, 13C NMR and mass spectrometry.
