Protective Effects of Quercetin on Doxorubicin-Induced Hepatotoxicity in male Rats

Objectives: Doxorubicin drug is an anti-cancer substance, but unwanted side effects result from its use.Dox is reported to cause cardio toxicity, nephrotoxicity, and hepatotoxicity. This toxicity seems to be due to the generation of reactive oxygen species (ROS) including hydroxyl radicals and superoxide anions thus leading to lipid peroxidation and tissue damage. The objective of this research was to study the possibility of whether Quercetin has the ability to reduce the harmful effects of the drug on vital organ like liver.Quercetin belongs to a group of plant pigments called flavonoids that give many fruits, flowers, and vegetables their colors. Flavonoids, such as quercetin, are antioxidants. Materials Methods: Male rats weighting 200-220 g were divided into 4 groups: Control, DMSO was given orally by gavage for 14 days.Quercetin (20 mg/kg) was given orally by gavage for 14 days.Doxorubicin was injectedintraperitonealy(25 mg/kg) on day 12th, 13th and 14th during the experiment.Quercetin-doxorubicin group;Quercetin(20 mg/kg/day oral) was given to animals for 14 days and Dox was administered (25 mg/kg; i.p.)on 12th, 13th and 14th day. After completion of the experiment animals were dissected and samples of livers to prepare textile sections for routinehistopathological examination were taken. The histopathological examination in doxorubicin-treated rats revealed some deterioration in liver functions and histopathology. Thesedeteriorations were prevented by pretreatment with Quercetin. Also liver samples in each group was homogenizedfor PBS cool for 1 minute with rpm10000 and then centrifuged for 2 minutes in a refrigerated centrifuge, and samples were taken for enzyme ALT, AST, ALP.Results Conclusion: Quercetin caused significant reduce of ALT,AST,ALP.activity signs compared to Doxorubicin group.(Pvalue 0.05). In conclusion, the use of Quercetin with toxic anticancer agent DOX may decrease the bad side effects of that drug. This study indicated that Quercetin has a protective effect on doxorubicin-induced hepatotoxicity in rats.
