    Lectin Histochemical Determination of Some Glycoconjugates in the Testes of Curly Dwarf Cochin (Gallus gallus)

  • Abstract
    This study was aimed to determine some glycoconjugates in testis of Curly Dwarf Cochin (Gallus gallus) through lectin histochemical method. The sections were incubated with the Horseradish peroxidase (HRP)-conjugated Canavalia ensiformis agglutinin (Con A), Glycine max agglutinin (SBA), Ulex europaeus agglutinin (UEA-I), Arachis hypogaea agglutinin (PNA) and Triticum vulgaris agglutinin (WGA) lectins in order to determine the glycoconjugate content. Lectin histochemical results indicate that spermatagonium and Leydig cells reacted very strongly with Con A lectin, but the peritubular cells of the lamina propria did not react with this lectin. A moderate reaction to PNA and very strong reaction to WGA were observed in primary spermatocytes. It was noticed that it reacted very strongly with UEA-I lectin while SBA lectin did not bind to the basal lamina. A moderate reaction to Con A, SBA and UEA-I, and weak reaction to PNA and WGA were detected in Sertoli cells. In conclusion, it has been determined that glycoconjugates in secondary spermatocytes and early spermatids contain less α-D-Mannose (α-D-Man), α-D-Glucose (α-D-Glc) and α-L-Fucose (α-L-Fuc) than the other stages, and that glycoconjugates in all spetmatogenic cells contain a large amount of sialic acid. However glycoconjugate in Leydig cells had all researched sugar residues.
