  • Abstract
    We prepared PEI-As2O3/Mn0.5Zn0.5Fe2O4 magnetic complex as drug and pHsp-HSV-TK gene vector, at the same time it could induce the gene expressed by heat. Then the MTT assay, hemolysis test, the mouse of LD50 test and micronucleus (MN) test were presented to evaluate the toxicity of it in vitro and in vivo . The therapeutic effects on cultured HepG2 cells treated by thermochemotherapy and pHsp70-HSV-TK of nanosized PEI-As2O3/Mn0.5Zn0.5Fe2O4 complex were evaluated by MTT test and flow cytometry. The experiments showed that the toxicity of the material on mouse fibroblast (L-929) cell lines was between 0~1 degree; It has no hemolysis activity; Its LD50 arrived at 3.449 g/kg; Micronucleus test showed it has no genotoxic effects either. The therapeutic results in vitro showed that thermochemotherapy combined with gene therapy of PEI-As2O3/Mn0.5Zn0.5Fe2O4 complex could significantly inhibit cellular growth and obviously induce cellular apoptosis. Furthermore, the therapeutic results outweigh that of single As2O3 group and Mn0.5Zn0.5Fe2O4 group.
