  • Abstract
    The first mapping of intracellular calcium transients in perfused rabbit hearts is described. Hearts were loaded with fluo 3 AM by coronary perfusion. Fluorescence was excited with a 488 nm steered laser beam, collected with a concave reflector, passed through a 515 nm longpass filter and recorded with a photomultiplier tube. Calcium transients had shapes similar to that reported in isolated myocytes, propagated anisotropically consistent with action potential propagation, and were sensitive to activation rate and verapamil. It is concluded that multisite mapping of calcium transients in hearts may provide insights into roles of calcium in abnormal heart activity
