Abstract :
High performance liquid chromatography (HPLC) method was coupled with pulsed amperometric detector, a kind of electrochemical detector by combining with post-column sodium hydroxide solution in order to analyze glycosides. Using this method, the contents of amygdalin, baicalin, naringin, and hesperidin in TCMs were determined with isocratic elution of 13% acetonitrile with 10 mM sodium phosphate buffer (pH 6.9). The coefficient of linear regression was 0.9997-0.9999 for concentrations between 1 and 100 μg/mL. The precision (RSD %) was less than 8.89% in Chong-Phyo-Tang and Run-Chang-Tang. This method showed high selectivity for analyzing glycosides without any interfering peaks. Therefore, glycosides in TCMs were conveniently analyzed without any pretreatment steps.
