Abstract :
Fluorescence microscopy is an important optical microscopic observation apparatus used for the research of cytobiology and cytogenetics. By using microscopic photography, digital CCD imaging instrument and other devices, researchers can observe and capture images of FISH, GISH, and Green fluorescent protein (GFP), etc. In practice, however, in order to get good experimental results with a clear background and distinctive hybridization signal for GISH in Triticeae, it is necessary to improve GISH image processing and the resolution. In this paper the main points for improving GISH technology and image effects in Triticeae are discussed, such as Chromosome preparation, the technical requirements of the probe, application of hybrid reaction promoter, and fluorescence microscopy observation, photography and image processing, etc.
