An indirect competitive enzyme-linked immunosorbent assay was developed for measurement of 2,2-bis-(4-chlorophenyl) acetic acid (DDA). Organochlorine pesticide 1,1,1-trichloro-2,2-bis- ( p-chlorophenyl ) ethane (DDT) is a persistent and broad toxic organochlorine insecticide. DDT is slowly degraded in tissues mainly to DDA. DDA was coupled to BSA and OVA by carbodiimide reaction. DDA-BSA as immunogen were injected in BALB/c mice. Titres of the antisera against DDA were determinated using DDA-OVA as coating ligand by ELISA method. The spleen cells of immunized mice were fused with Sp2/0 cells. After cloning, 23 hybridoma cell strains stably produced anti-DDA monoclonal antibody were obtained. The monoclonal antibody was produced by the mouse ascites method. The indirect competitive ELISA for DDA in shellfish was established. Under optimal condition, the detection limit of DDA was 1.90ng/mL and the coefficient of variation was 0.10-5.17%, the average recovery was 120.9-142.5%.
