Abstract
In vivo cell tracking is an emerging field and a promising technique for tracking stem cells, metastasis and inflammation. The aim of this study was to find optimal US parameters for ultrasound contrast agent-mediated SPIO-labeling of endothelial cells for MRI tracking. The optimal ultrasound parameters were 40 kPa at 1 MHz, 10,000 cycles, 20 Hz pulse repetition frequency, applied 5 minutes after SPIO addition. This gave a ~7 times increase of SPIO uptake in endothelial cells in vitro compared to the control with 90% cell viability. This technique could therefore be safely used for labeling of endothelial cells with SPIO for MRI tracking.
