Investigation on beet mosaic virus in Khorasan Province

Beet mosaic virus (UtMV) is a member of potyvirus genus, is transmitted readily by sap inoculation and aphid in a non- persistent manner. Initial symptoms appear as chlorotic spots on young leaves. The spots are more or less circular, often with sharply defined margins. In this study DASELISA was used to detect BtMV in sugar beet leaves collected from different fields in the main sugar beet growing areas of Khorasan Province. Isolates from Nyshabor regions especially Gadamgha were used for host range study, virus purification and dot immunobinding assay. To determine molecular weight of virus coat protein, SDS-PAGE and western blotting were conducted, and the antiserum to BtMV CP reacted strongly with CP of the BtMV isolate and a single protein band of 34700 Da was revealed on nitrocellulose membrane.
